Resumen del proyecto: Functional disorders affecting the signaling pathway of lysophosphatidic acid in animal models result in defects in neurogenesis as well as a greater propensity to develop an addictive behavior. The role of this pathway in humans is, however, largely unexplored. A review of data generated in GWAS for smoking and other behavioral disorders suggests that some common markers of the human form of the gene family could be capturing LPAR risk factors to these diseases. The in silico analysis of the genomic environments of the candidate genes revealed clusters of different SNPs located in the vicinity of the LPAR2 and LPAR1 which can be controlling their genes transcript levels and, therefore, condition the relative risk of people develop an addictive behavior. We have selected a total of 25 SNPs that are well TagSNPs positive eQTL studies or related phenotypes previously associated with but that did not make the cut off necessary to pass the Bonferroni correction in genome studies. Power models suggest that with some allelic frequencies between 0.15 and 0.49, we have a 80% power to find Odds ratios> 1.43 using 500 cases and 500 controls from the general population.
