Título: Identification of non-invasive biomarkers of progression of liver damage and useful as targets for the treatment of aged patients

Resumen del proyecto: As part of a broader project aimed at identifying new biomarkers that allow detect the progression of liver damage the two main types of hepatic tumors: hepatocarcinoma (HCC) and cholangiocarcinoma (CCA), one of the objectives of this Project is to determine metabolites in serum samples of patients with distal CCA (dCCA) and pancreas ductal adenocarcinoma (PDAC) since, when these tumors are localized in the pancreatic head, the diagnosis can be difficult. To this end, the metabolomic profile will be determined by liquid chromatography-mass spectrometry (UHPLC/MS) in three platforms (polars, lipids, and aminoacids). In particular, the service includes the extraction, measurement and analysis of metabolites (bile acids, non-esterified fatty acids, phospholipids, sterols, sphingolipids, glycerolipids, etc) in serum samples of patients with dCCA, PDCA and controls. Moreover, samples from patients with benign liver (esteatohepatitis) and pancreas (chronic pancreatitis, cysts, etc) will be also included and development of statistical evaluations among groups will be carried out. Metabolomic analyses will be carried out by One Way Liver S.L. (OWL), a biotechnology company in the field of metabolomics with pioneering diagnostics products and unique R&D services for the scientific community, and the conditions have already been established. The aim is to identify specific biomarkers that allow a differential diagnosis between both types of tumors. In addition to the request of serum samples to the Biobanks Network DNA National Bank Carlos III (University of Salamanca), similar samples have been requested to the biobanco vasco del Instituto de Investigación Sanitaria Biodonostia de San Sebastián (Dr. Luis Bujanda). We aim to achieve about 50-60 samples of each type for a first analysis and them to obtain more samples for a validation phase.
