Título: Identification of metastatic colorectal cancer mutations by next generation sequencing approaches. Paired quantification studies in tumor circulating DNA.
IP: Dr. Luis Muñoz
Resumen del proyecto: The study will undertake the ultrasensitive detection of circulating tumor DNA (ctDNA) in plasma of patients colorectal cancer evaluating the detectability of specific mutations (liquid biopsy). To this aim, firstly we will identify the specific mutation in the tumor sample for each patient by Next generation sequencing (NGS). Thereafter, a highly sensitive qPCR-based method will be set-up and standardized to detect and quantify each mutation in the plasma ctDNA of the patients to test the correspondence with the previously found tumor markers. Finally, the analysis of the correlations between the biomarkers identified (in the primary tumor and ctDNA samples) with the clinical-biological characteristics and the clinical-pathological parameters of each patient will allow to determine the effective clinical translational value of the detection of mutation markers and the correspondence of such markers with the disease fate and treatment response. The proposal success will have a great translational value to the clinical practice to improve patient care, employing a non-invasive procedure to provide highly innovative predictive, preventive and personalized medicine
